Restriction site variation in the nuclear 18S–25S ribosomal RNA genes (rDNA) was analyzed hierarchically in a species complex in the fern genusPolystichum. Two distinct rDNA repeat types were present in all individuals ofPolystichum examined. No variation was detected among individuals within a population ofP. munitum, among populations ofP. munitum orP. imbricans, or among the six diploid species ofPolystichum from North America, including the circumborealP. lonchitis. The identity of rDNA repeats across all six North American species ofPolystichum may reflect an overall similarity of the nuclear genomes of these species, an observation supported by isozyme data as well. However, this nuclear similarity contrasts sharply with the highly divergent chloroplast genomes of these six species. The conservative nature of the rDNA inPolystichum also is in contrast to the much more variable rDNAs of most angiosperms investigated. Perhaps the tempo and mode of evolution of rDNA in ferns differ from those of angiosperms; however, the data base for fern rDNA is very small. Furthermore, the number of repeat types per individual is consistent with a diploid, rather than polyploid, condition despite the high chromosome number (n = 41) of these plants, although homogenization of multiple, divergent rRNA genes cannot be disproven.  相似文献

The abundance of Cynodon dactylon was recorded in 50 plots presenting different geomorphological conditions and along a transect of 16 500 2×2 cm contiguous quadrats within a small basin 330 m in length, in the granite pediment of the Sierra de Guadarrama (Central Spain). Soil analyses were undertaken on samples from the 50 plots and the soil information matrix obtained was analysed using Principal Components Analysis and Discriminant Analysis. Results showed that in the Mediterranean pastures Cynodon dactylon was restricted to deep, well developed soils with relatively high values of cations, conductivity and organic matter, and could be used as an indicator of such soil conditions in these grasslands. The spatial pattern of the species was analysed using New Local Variances which revealed the existence of a small-scale pattern, ranging from 8 to 18 cm, present at all levels of the analysis. The size of small-scale patterns was positively correlated with species abundance and was interpreted as a relation between plant vigour and favourable soil conditions.I would like to thank Professor González Bernáldez for his interest and encouragement.  相似文献

Summary Equivalent-circuit impedance analysis experiments were performed on the urinary bladders of freshwater turtles in order to quantify membrane ionic conductances and areas, and to investigate how changes in these parameters are associated with changes in the rate of proton secretion in this tissue. In all experiments, sodium reabsorption was inhibited thereby unmasking the electrogenic proton secretion process. We report the following: (1) transepithelial impedance is represented exceptionally well by a simple equivalent-circuit model, which results in estimates of the apical and basolateral membrane ionic conductances and capacitances; (2) when sodium transport is inhibited with mucosal amiloride and serosal ouabain, the apical and basolateral membrane conductances and capacitances exhibit a continual decline with time; (3) this decline in the membrane parameters is most likely caused by subtle time-dependent changes in cell volume, resulting in changes in the areas of the apical and basolateral membranes; (4) stable membrane parameters are obtained if the tissue is not treated with ouabain, and if the oncotic pressure of the serosal solution is increased by the addition of 2% albumin; (5) inhibition of proton secretion using acetazolamide in CO2 and HCO 3 -free bathing solutions results in a decrease in the area of the apical membrane, with no significant change in its specific conductance; (6) stimulation of proton transport with CO2 and HCO 3 -containing serosal solution results in an increase in the apical membrane area and specific conductance. These results show that our methods can be used to measure changes in the membrane electrophysiological parameters that are related to changes in the rate of proton transport. Notably, they can be used to quantify in the live tissue, changes in membrane area resulting from changes in the net rates of endocytosis and exocytosis which are postulated to be intimately involved in the regulation of proton transport.  相似文献

The submerged vegetation of Lake Kariba is described in relation to degree of slope (lake morphometry), depth and light transparency. The direct gradient analysis technique — canonical correspondence analysis and the TWINSPAN classification programs were used to analyse the data set. The western end of the lake with low transparency has a low species diversity (with Vallisneria aethiopica dominating). Species diversity increases with increased transparency in the other parts of the lake. The classification revealed monospecific communities for all species as well as mixed communities with Lagarosiphon as the associate species with the broadest distribution. The ordination revealed a first axis strongly related to depth and transparency gradients and the second axis related to slope. Vallisneria aethiopica has a growth form adapted to grow in shallow areas subjected to wave action. Potamogeton octandrus also growing in shallow areas is restricted to sheltered areas. Lagarosiphon ilicifolius precluded from the shallow exposed areas grows at medium depth but has a growth form adapted to tap light at the surface. Najas pectinata and Ceratophyllum demersum are adapted to grow in deep water.Abbreviations CCA Canonical correspondence analysis  相似文献

In this article a test for the comparison of K groups of independent profiles is suggested. It is based on rank tests, in which the criterion is obtained by using a suitably chosen ordinal classification function. This function represents a specific view on the profiles. The test, which is easy to use, can be regarded as a generalisation of other well known tests and can be applied within a variety of situations. The relationships to other procedures for the analysis of profiles are described. Some properties are discussed.  相似文献

Summary Six different statistical methods for comparing limiting dilution assays were evaluated, using both real data and a power analysis of simulated data. Simulated data consisted of a series of 12 dilutions for two treatment groups with 24 cultures per dilution and 1,000 independent replications of each experiment. Data within each replication were generated by Monte Carlo simulation, based on a probability model of the experiment. Analyses of the simulated data revealed that the type I error rates for the six methods differed substantially, with only likelihood ratio and Taswell's weighted mean methods approximating the nominal 5% significance level. Of the six methods, likelihood ratio and Taswell's minimum Chi-square exhibited the best power (least probability of type II errors). Taswell's weighted mean test yielded acceptable type I and type II error rates, whereas the regression method was judged unacceptable for scientific work.  相似文献
